Spence is a Chattanooga native and the oldest of six children, all of whom are college graduates. He is married to Dr. Cynthia Neal Spence, a professor at Spelman College in Atlanta. They have been married since 1980 and have two daughters, Christie (Ph.D.) and Courtnee (MBA).
